Can I use the USB Port on my DS220+ to load Photos and videos from the USB to my NAS? If so will I have a choice as to what folder on the NAS the Photos go to?
Yes and yes. Use File Station and transfer what you want where you want.

Media indexing works by you defining which locations to index, and selecting what type of media they contain: photo, music, video.

You haven’t mentioned it but if you are thinking about accessing the photos using Synology Photos then you must transfer them to the right folders.
